The Supreme Court decision upholding President Barack Obama's health care overhaul affects nearly every American, because the law itself is so vast. A look at what it does and what's to come:

WHAT DOES THE RULING MEAN TO ME?

The law tells almost everyone they must have health coverage and guarantees it will be available even if you are already ill or need hugely expensive care. It helps lots of people, not just the poor but many upper-income families, as well, afford coverage. And it requires insurers to provide certain basic benefits ? like free preventive care.

WHAT THE JUSTICES SAID

The high court upheld almost all of the law, including the most disputed part: the mandate that virtually all Americans have health insurance or pay a penalty. The court said that penalty is essentially a tax, and that's why the government has the power to impose it.

The ruling limited the law's plan to expand the Medicaid insurance program for the poor, a joint effort of the federal government and states. It says the U.S. government cannot withhold a state's entire Medicaid allotment if it doesn't participate in the expansion.

Chief Justice John Roberts sided with the court's four liberal justices ? Stephen Breyer, Ruth Bader Ginsburg, Elena Kagan and Sonia Sotomayor ? to form the 5-4 majority.

The court upheld Obama's signature legislative achievement. Final word from the court amplifies the most polarizing issue of his re-election campaign against Republican Mitt Romney.

GOP lawmakers and Romney have promised to repeal the law if they are in power after the November election.

WHAT NOW?

The 2010 health care law will keep taking effect. It's expected to bring coverage to about 30 million uninsured people. Overall, more than 9 in 10 eligible Americans will be covered.

Some parts are already in effect: Young adults can stay on their parents' insurance up to age 26. Insurers can't deny coverage to children with health problems. Limits on how much policies will pay out to each person over a lifetime are eliminated. Hundreds of older people already are saving money through improved Medicare prescription benefits. And co-payments for preventive care for all ages have been eliminated.

WHAT'S NEXT?

Starting in 2014, almost everyone will be required to be insured or pay a penalty. There are subsidies to help people who can't afford coverage. Most employers will face penalties if they don't offer coverage for their workers. Newly created insurance markets will make it easier for individuals and small businesses to buy affordable coverage. And Medicaid will be expanded to cover more low-income people.

Insurers will be prohibited from denying coverage to people with medical problems or charging those people more. They won't be able to charge women more, either. During the transition to 2014, a special program for people with pre-existing health problems helps these people get coverage.

An assortment of tax increases, health industry fees and Medicare cuts will help pay for the changes.

STILL, NOT EVERYONE WILL BE COVERED

An estimated 26 million people will remain without coverage once the law is fully implemented, including illegal immigrants, people who don't sign up and choose to face the penalties instead, and those who can't afford it even with the subsidies. That number could be higher, depending on whether any states refuse the Medicaid expansion.

THE TAXING TRUTH

When the law was before Congress, Obama and Democrats avoided calling its penalty for going uninsured a "tax." But the administration argued before the Supreme Court that the law was constitutional as a federal tax. The court rejected two other Obama administration arguments for the law but accepted the tax assertion.

Not everyone without insurance will be docked. By 2016, about 4 million people will pay the penalty to the Internal Revenue Service, the Congressional Budget Office has estimated. They would pay $695 for each uninsured adult or 2.5 percent of family income, up to $12,500 a year.

The IRS can't prosecute violators or place liens against them, however. Its only enforcement option may be withholding money from refunds.

WHAT ARE REPUBLICANS SAYING?

"Obamacare was bad law yesterday. It's bad law today," Romney said after the ruling.

The Republican-led House already has voted for repeal but can't push it forward so long as Obama's in the White House and Democrats lead the Senate ? making the November elections crucial.

Senate Republican leader Mitch McConnell called the court decision "a fresh start on the road to repeal."

OBAMA'S REACTION

Obama says the decision upholds the fundamental principle that in the wealthiest nation on earth, no one should be ruined financially by an illness or accident.

He called it "a victory for people all over this country whose lives will be more secure because of this law."

ABOUT THAT MANDATE

Some parts of the law have proven popular. But the insurance mandate is widely disliked.

Each time The Associated Press has asked in polls, more than 8 in 10 Americans have said the government should not have the right to require everyone to buy health insurance.

The public also has tilted against the law as a whole over the two years since it was passed. About half opposed it and a third were in favor in an AP-GfK poll shortly before the Supreme Court ruled.

Astronomers discover an 'odd couple' of planets

The Kepler spacecraft has detected a pair of extrasolar planets?with orbits so close that at times the larger planet looms more than twice the size of the full moon in the second planet's night sky.

Astronomers using NASA's Kepler planet-hunting spacecraft have discovered an Oscar and Felix among extrasolar planets ? an odd couple with orbits so close to each other that at times the larger planet looms more than twice as large as the full moon in the second planet's night sky.

The duo orbits a star 750 light-years from Earth, one sun among some 150,000 stars that Kepler monitors in the hunt for Earth-like planets orbiting sun-like stars at Earth-like distances.

This pair, however, occupies a system unlike any detected so far.

Individually, each planet falls into a class detected elsewhere. The smaller, inner planet, Kepler-36b, has about 4.5 times Earth's mass and is about 1.5 times as large. It's a rocky orb with an estimated 30 percent of its mass in the form of iron.

Its larger, outer companion Kepler-36c, is more like a mini Neptune. It boasts eight times Earth's mass, but has a density suggesting a gas-giant wannabe with a "substantial" atmosphere of hydrogen and helium, according to the team making the discovery.

?But the planets' two orbits are separated by an average of only about 1.2 million miles ? cosmic overcrowding by the standards of Earth's planetary neighborhood. Mars' orbit is about 49 million miles outside Earth's path, while Venus' orbital path lies roughly 27 million miles inside Earth's.

Two planets of such dramatically different compositions and such tightly packed orbits represent "an exciting system," says Eric Agol, an astronomer at the University of Washington in Seattle and one of the two lead authors on the formal report of the finding published Thursday on Sciencexpress, an online adjunct to the journal Science.

The planets reach their closest encounters ? less than 1.2 million miles apart ? when they line up on the same side of the star once every 97 days on average, in a configuration known as an inferior conjunction.

Life is highly unlikely on these orbs. They are so close to their host star ? with orbits of 13.8 days for the inner planet and 16.2 days for the outer planet ? that temperatures hover between 1,200 and 1,300 degrees F.

And they are likely to get worse. Kepler-36b is from 2 billion to 3 billion years older than the sun, the team estimates. The star is a so-called sub-giant, meaning it's burned up the hydrogen in its core and is headed down a path that will turn it into a red giant. The star's core is contracting, allowing it to burn hotter and brighter ? hot enough to begin fusing the hydrogen outside the core. Like the sun's end-of-life sequence, Kepler-36's fiery atmosphere will expand much farther into space than it does now, engulfing the odd couple.

As planet-hunters uncover such seemingly bizarre systems, "we're constantly being surprised," says Joshua Carter, the other lead author and a Hubble Fellow at the Harvard-Smithsonian Center for Astrophysics in Cambridge, Mass. "But we're just surprised because we're basing our preconceptions on the solar system."

Planets in our solar system tend to have organized into two groups ? the rocky inner planets and the giant, gaseous outer planets. The rocky planets formed close to the sun, where any gas the planets' gravity couldn't capture and retain was vaporized. Beyond the so-called snow line, between Mars and Jupiter, temperatures were cool enough for gas to remain a gas, as well as to shift from gas, to liquid, and finally ice.

With orbits so close and so close to their star, but with such a stark contrast in density (an eight-fold difference) and composition, the odd couple would seem to require a different explanation, the researchers say. It's possible that the rocky inner planet formed inside the frost line, while the mini-Neptune formed outside and migrated inward over time, Dr. Agol acknowledges. Migration seems to be the leading explanation for the large number of gas giants ? so-called hot Jupiters and Neptunes ? planet hunters have uncovered during the past two decades.

Migration explanations could account for the system the team detected, Dr. Carter agrees. But such an explanation "would have to simultaneously account for the closeness and the disparity in density."

One possible explanation Agol says he finds appealing is a variation of migration where both planets began as gas-giant wannabes beyond the snow line, then migrated in with time and settled into the orbits the team identified. But the inner planet may not have had enough mass to allow its gravity to retain its atmosphere so close to its stellar furnace, while the larger of the two was just far enough away from the star and just big enough for its gravity to keep a grip on an atmosphere.

"One of the most exciting things about this system is that maybe these planets are straddling a line" that determines whether planets, which may have started out with comparable masses when they first formed, will retain or lose their atmospheres, he says.

The team found the system as it analyzed data released early in Kepler?s mission. The craft launched in March 2009 and is now in an extended phase of its mission.

Kepler detects candidate planets by constantly watching the 150,000 stars in its field of view for the slight dimming a planet would impart as it passes in front of its host star. That dimming revealed the presence of the larger of the two planets orbiting Kepler-36. But irregularities in the timing of the dimming suggested that the bigger planet might have a sibling.

During the past year or so, as Kepler gather more data on the system, Carter and Agol have been perfecting a mathematical tool for analyzing such irregularities to tease out information on both planets from the subtle dimming effects Kepler picks up.

Kepler's exquisite measurements of key properties of the host star allowed the team to use the transits to determine the size of each planet, while the orbital periods allowed the team to estimate the masses of the planets ? calculations that become more precise when the gravitation interaction between the two planets is included. Armed with these key traits, the researchers estimated the density for each planet, allowing them to distinguish the difference between the mini Neptune and the rocky super Earth.

Court throws out FCC penalties for cursing, nudity

WASHINGTON (AP) ? Broadcasters anticipating a major constitutional ruling on the government's authority to regulate what can be shown and said on the airwaves instead won only the smallest of Supreme Court victories Thursday.

The justices unanimously threw out fines and other penalties against Fox and ABC television stations that violated the Federal Communications Commission policy regulating curse words and nudity on television airwaves.

Forgoing a broader constitutional ruling, however, the court concluded only that broadcasters could not have known in advance that obscenities uttered during awards show programs on Fox stations and a brief display of nudity on an episode of ABC's "NYPD Blue" could give rise to penalties. ABC and 45 affiliates had been hit with proposed fines totaling nearly $1.24 million.

Broadcasters had argued that the revolution in technology that has brought the Internet, satellite television and cable has made the rules themselves obsolete. The regulations apply only to broadcast channels.

The justices said the FCC is free to revise its indecency policy, which is intended to keep the airwaves free of objectionable material during the hours when children are likely to be watching.

The agency's chairman, Julius Genachowski, said the ruling "appears to be narrowly limited to procedural issues related to actions taken a number of years ago. Consistent with vital First Amendment principles, the FCC will carry out Congress's directive to protect young TV viewers."

It was the second time the court has confronted, but not ruled conclusively on the FCC's policy on isolated expletives. Justice Anthony Kennedy said in his opinion for the court that "it is unnecessary for the court to address the constitutionality of the current policy."

The narrow decision, coupled with the more than five months that elapsed between the argument in January and Thursday's decision, could mean that the justices struggled and failed to reach agreement on a broader outcome.

Broadcasters argue that viewers now have many options, unlike the handful of channels they had available in the 1960s and 1970s when the court last weighed in on indecency on the airwaves. In many cases, viewers don't even know when they are switching between the older broadcast channels and cable.

Still, the regulated broadcast channels provide what the government has called a safe haven of milder programming, and those channels remain dominant, even in the Internet age, the administration argued.

Paul Smith, a First Amendment expert and partner with the Jenner and Block law firm in Washington, said the court should expect more challenges until it rules definitively.

"The Supreme Court decided to punt on the opportunity to issue a broad ruling on the constitutionality of the FCC indecency policy. The issue will be raised again as broadcasters will continue to try to grapple with the FCC's vague and inconsistent enforcement regime," said Smith, who wrote a brief supporting the broadcasters.

The case arose from a change in the FCC's long-standing policy on curse words.

For many years, the agency did not take action against broadcasters for one-time uses of curse words. But after several awards shows with cursing celebrities in 2002 and 2003, the FCC toughened its policy after it concluded that a one-free-expletive rule did not make sense in the context of keeping the airwaves free of indecency when children are likely to be watching television.

But Kennedy, in the ruling throwing out the fines, said the commission did not adequately explain that under the new policy "a fleeting expletive or a brief shot of nudity could be actionably indecent."

The stepped-up indecency enforcement, which included issuing record fines for violations, also was spurred in part by widespread outrage following Janet Jackson's breast-baring performance during the 2004 Super Bowl halftime show on CBS.

That incident and the FCC's proposed fine of $550,000 are not part of the current case. The government has an appeal pending of a lower court ruling that threw out the fine in that case.

The 2004 Super Bowl took place before the FCC later that year laid out its new policy and the possibility of fines for even one-time utterances of certain words.

Tim Winter, president of the Parents Television Council, said he read the new decision as a "green light" for the FCC to rule against broadcasters in the many pending complaints of indecent material that aired after the FCC explained its new policy.

"Once again the Supreme Court has ruled against the networks in their yearslong campaign to obliterate broadcast decency standards," Winter said.

The material at issue in Thursday's decision included the isolated use of expletives as well as fines against broadcasters who showed a woman's nude buttocks on a 2003 episode of "NYPD Blue."

In December 2002, singer Cher used the phrase "F--- 'em" during the Billboard Music Awards show on the Fox television network. A month later, U2 lead singer Bono uttered the phrase "f------ brilliant" during NBC's broadcast of the Golden Globes awards show. During the December 2003 Billboard awards show on Fox, reality show star Nicole Richie said, "Have you ever tried to get cow s--- out of a Prada purse? It's not so f------simple."

But the challenge went beyond just the penalties for the use of fleeting expletives.

The broadcasters wanted the court to free them from all regulation of content around the clock. The court's 1978 Pacifica decision upheld the FCC's reprimand of a New York radio station for airing a George Carlin monologue containing a 12-minute string of expletives in the middle of the afternoon.

Justice Ruth Bader Ginsburg said in a brief opinion that she would have overturned the Pacifica ruling, which she called wrong even when it was decided. Justice Sonia Sotomayor did not take part in the current case because she was involved in an earlier version while sitting as an appeals court judge in New York.

The case is FCC v. Fox, 10-1293.

The ancient reserves of methane gas seeping from the melting Arctic ice cap told Jeff Chanton and fellow researchers what they already knew: As the permafrost thaws, there is a release of methane, a powerful greenhouse gas that causes climate warming.

The trick was figuring out how much, said Chanton, the John W. Winchester Professor of Oceanography at Florida State University.

The four-member team ? whose findings were published in the respected journal Nature Geoscience ? documented a large number of gas seep sites in the Arctic where permafrost is thawing and glaciers receding (they found 77 previously undocumented seep sites, comprising 150,000 vents to the atmosphere). Until recently, the cryosphere (frozen soil and ice) has served to plug or block these vents. But thawing conditions have allowed the conduits to open, and deep geologic methane now escapes.

The team studied the link between natural gas seepage and the melting ice cap, using aerial photos and field data to figure out the number ? and location ? of seep holes.

So, here's the rub: The more the ice cap melts, the more methane is released into the atmosphere ? and the more the climate warms.

Why should this matter to you?

People who live in coastal areas in Florida could be directly affected, said Chanton, who analyzed the methane and dated it to more than 40,000 years old.

All this seeping methane causes more melting ice, Chanton said, which causes sea levels to rise and could affect coastal real estate values ? sooner rather than later.

How soon?

Possibly over the next 50 to 100 years, Chanton said.

"Methane is a very strong greenhouse gas that's grown three times faster than carbon dioxide since the industrial era," Chanton said. "As the Arctic warms, the ice caps melt and the fissures open, so methane escapes and causes more warming."

This phenomenon causes sea levels to rise, which is particularly problematic in Florida:

"Along the flat Florida coastline, a 1-foot rise in sea level could cause anywhere from 10 to 100 feet of shoreline retreat ? erosion," Chanton said. "For us here in Florida, this is really important because we can expect the coast to recede."

That beach house, he warned, might be in peril: "It may not be there for your grandchildren."

If you're just taking down the decorations from your latest Google+ party, hang fire, it looks like there's more good news for the ever-burgeoning social starlet. It's just been announced that Google has developed an API that will make users' photos, posts and videos available on popular social reader Flipboard. Until now, the search giant's social network had been notable by its absence, but soon you'll be able to flick your way through your circles, just like your other streams. There's no indication as to when, but the new API shows Google isn't shy about spreading the love, even if it means treading on its own toes.

I submitted a character, you said we can't play both Antagonists and Protagonists, does that mean we can't have both a vampire and a townsperson?

Edit: I am also assuming standard vampire stuff? Sunlight is death, must drink blood, vomit up any type of human food, die from a stake to the heart, die if you cut the head off?
Are we doing the super powers or just the immortality(which I prefer myself)?

Colombia volcano Ruiz spews gas

Plumes of smoke and ash are rising from Colombia's Nevado del Ruiz volcano.

People living on its slopes said they had heard "strong, strange noises" coming from the summit of the 5,346m-high mountain on Friday and Saturday.

Officials said they would extend the orange alert, the second highest, for areas near the summit, which they had first declared almost three weeks ago.

More than 25,000 people died in a mudslide caused by Nevado del Ruiz in its last eruption in 1985.

Officials have distributed 30,000 face masks to residents living in areas affected by ash from the volcano.

Deadly mudslide

The airport in the nearby town of Manizales, in central Colombia, has been closed since the end of May, and will remain so until the volcano seizes to emit ash, officials said.

The Volcanic Observatory in Manizales said the plume of ash and gas had risen to a height of 2,000m (6,500ft) on Sunday.

Scientists at the observatory warned there was a risk of an eruption "within days or weeks".

One-hundred-and-fifty families have been evacuated from the banks of the rivers which flow down the volcano, as they were deemed to be most at risk.

In the 1985 eruption, the small town of Armero on the banks of the Lagunilla river was almost completely erased as summit glaciers and snow were melted by hot lava.

The water raced down the slopes carrying rocks, mud and vegetation in a destructive mudslide travelling at a speed of 60km/h (37mph).

Only a quarter of Armero's population survived.

China to launch woman into space

China is about to launch its fourth manned space mission.

It is sending a crew of three, including the nation's first female astronaut, to the orbiting Tiangong space lab.

Their Shenzhou-9 capsule is set to lift off from the Jiuquan spaceport on the edge of the Gobi desert at 18:37 local time (10:37 GMT; 1137 BST).

A Long March 2F rocket will put the astronauts on a path to dock with Tiangong in a couple of days' time.

They will then spend over a week living and working in orbit before returning to Earth.

The mission is commanded by Jing Haipeng, who is making his second spaceflight after participating in the Shenzhou-7 outing in 2008 - the mission that included China's first spacewalk.

Jing's flight engineers are both first-timers.

Liu Wang, a People's Liberation Army fighter pilot, has got his chance after spending 14 years in the China National Space Administration's astronaut corps.

Liu Yang, on the other hand, has emerged as China's first woman spacefarer after just two years of training.

Her role in the mission will be to run the medical experiments in orbit.

Shenzhou-9 follows on from the unmanned Shenzhou-8 venture last year that tested the technologies required to join a capsule to the Tiangong lab.

Those manoeuvres went very well and gave Chinese officials the confidence to send up humans.

When it arrives at Tiangong, the Shenzhou-9 craft is expected to make a fully automated docking, but there is a plan to try a manual docking later in the mission.

This would see the crew uncouple their vehicle from the lab, retreat to a defined distance and then command their ship to re-attach itself.

Liu Wang will take the lead in this activity. "We've done many simulations," he said during the pre-launch press conference.

"We've mastered the techniques and skills. China has first class technologies and astronauts, and therefore I'm confident we will fulfil the manual rendezvous."

Tiangong is the next step in a strategy that Beijing authorities hope will lead ultimately to the construction and operation of a large, permanently manned space station.

It is merely the prototype for the modules China expects to build and join in orbit. Mastering the rendezvous and docking procedures is central to this strategy.

At about 60 tonnes in mass, this proposed station would be considerably smaller than the 400-tonne international platform operated by the US, Russia, Europe, Canada and Japan, but its mere presence in the sky would nonetheless represent a remarkable achievement.

Concept drawings describe a core module weighing some 20-22 tonnes, flanked by two slightly smaller laboratory vessels.

Officials say it would be supplied by freighters in exactly the same way that robotic cargo ships keep the International Space Station (ISS) today stocked with fuel, food, water, air, and spare parts.

China is investing billions of dollars in its space programme. It has a strong space science effort under way, with two orbiting satellites having already been launched to the Moon. A third mission is expected to put a rover on the lunar surface.

The Asian country is also deploying its own satellite-navigation system known as BeiDou, or Compass.

Before leaving Earth, Lui Yang said the Shenzhou-9 mission would generate further pride in Chinese people.

"When I was a pilot I flew in the sky; now as an astronaut, I'm going into space. It's higher and it's farther," she said.

"I have a lot of tasks to fulfil, but besides these tasks I want to feel the unique environment in space and admire the views. I want to explore a beautiful Earth, a beautiful home.

"I want to record all my feelings and my work, to share with my friends, and my comrades and my future colleagues."

Riverside launching website for fitness and healthy living - Riverside ...

The website is part of the Start R.I.G.H.T. (Riverside Is Getting Healthy Together) Campaign, which is hosted and lead by the Riverside Community Health Foundation.? The Campaign is a citywide initiative to provide education and opportunities to improve the obesity rates of the residents of Riverside, specifically to reduce the obesity and overweight percentages of Riverside residents to less than 50% in five years.

The website focuses on three areas:?Live Right?, ?Eat Right?, and ?Move Right?.

Mayor Ron Loveridge, a known advocate for fitness, said, ?Fit Fresh & Fun is an important quality of life initiative intended to raise health and wellness awareness, knowledge and motivation of individuals and to highlight the many resources here to help residents be healthy.? This new website provides a menu of tools such as health and wellness education articles, and an event calendar full of local events to get people moving.?

The website will feature ?Find a Class? ? a listing of classes, teams, or groups whose mission it is to improve health and have fun; and ?Ask an Expert? - an interactive forum where residents can ask health questions of real, local experts.

Additionally, starting in September, the website will have a ?My Passport? feature where participants can log their health progress and actively participate in the Start R.I.G.H.T. campaign in a more personal way.

Future features will include a local restaurant guide that helps our community makes the best dining choices at local restaurants.

L'Oreal Creates Haircolor Tribes on Facebook, Pinterest

loreal-facebookL'Oreal has teamed up with marketing agency Campfire to encourage friendly competition and a sense of camaraderie among blondes, brunettes and redheads on Facebook, Pinterest and beauty blogs.

The campaign, which is for L'Oreal's Healthy Look Creme Gloss semi-permanent color, asks women to "Turn it Up," by identifing their hair color, becoming a part of their so-called hair color tribe and expressing their hair color identity by using new tools on its Facebook page .

Merrin McCormick, associate creative director at Campfire, says L'Oreal was not getting the traction it wanted with its creme gloss product in the U.S. and wanted to find a way to engage its audience and create evangelists for the brand. Thus the concept of the Hair Color Tribe was born as it is a fun and shareable concept, she said.

Because Healthy Look Creme?Gloss semi-permanent color enhances natural hair?color rather than altering it completely, McCormick says, "It's about embracing who you are. You really love your hair color and this product doesn't have a drastic change - it only makes your hair a few shades lighter or darker."

One main component of embracing hair color is L'Oreal's 2012 Hair Color Census. It includes an introductory video and questions about personality and beliefs about hair color, like, "What haircolor do you believe is most trustworthy?" and "What haircolor do you believe is the most spontaneous?"

L'Oreal will compile the results for ongoing marketing and PR efforts, according to Campfire. That will include graphics and a second video, which will appear ongoing throughout the campaign. McCormick expects it will wrap up in August. It launched about a week ago.

The campaign will eventually extend to Pinterest, in which bloggers will be encouraged to?create "Turn It Up" inspiration boards. In addition, "Pin it" buttons throughout the?Facebook app will allow users to integrate content via their Pinterest boards.

Pinterest content will appear when L'Oreal has infographics from the Census as a way to refresh the campaign, McCormick added.

Additional content in the Facebook app includes: content from beauty?bloggers discussing what their color identities mean to them and documenting?their color through 28 washes; a cover photo tool to help?Facebook Timeline users express their color identity, as well as tools to?create posters and desktop wallpapers; coupons, offers and contests; a shade guide and product?tutorials; and a before and after photo gallery.

L'Oreal has also tapped "Hair Color Ambassadors" to represent each shade. They will talk about their hair color and encourage consumers to check out Facebook and their own blogs, McCormick adds.

The intention is also to take away some of the fear of at-home hair color. "It's more about adjusting to change," McCormick said.

A study led by Vanderbilt-Ingram Cancer Center (VICC) investigators has identified a gene expression pattern that may explain why chemotherapy prior to surgery isn't effective against some tumors and suggests new therapy options for patients with specific subtypes of breast cancer.

The study by lead author Justin Balko, Pharm.D., Ph.D., was published online June 10, 2012 in Nature Medicine in advance of print publication. Balko is a postdoctoral fellow in the laboratory of Carlos L. Arteaga, M.D., associate director for Clinical Research and director of the Breast Cancer Program at VICC, who led the study.

About 30 percent of breast cancer patients have a pathological complete response when chemotherapy is used to shrink tumors prior to surgery. However, many patients still have residual cancer in the breast after neoadjuvant chemotherapy (NAC) is completed. These patients are at a higher risk of cancer recurrence and death.

The investigators suspected that profiling tumors after neoadjuvant chemotherapy would identify genes associated with resistance to this form of treatment. They studied gene expression patterns in 49 breast tumors obtained during surgery after four months of NAC.

They identified and analyzed specific groups of genes associated with high-grade, chemotherapy-resistant tumors, labeling their 244 unique genes the CLUSTER signature, and combined this panel with previously identified gene signatures to search for distinctive patterns of behavior.

The investigators found that low concentrations of dual specificity protein phosphatase 4 (DUSP4) is strongly correlated with faster tumor cell growth following neoadjuvant chemotherapy. Low DUSP4 was also correlated with a type of breast cancer known as basal-like breast cancer (BLBC). DUSP4 promoter methylation and gene expression patterns of Ras-ERK pathway activation were also higher in BLBC relative to other breast cancer subtypes.

When DUSP4 was present, chemotherapy was effective against cancer cells, whereas when DUSP4 was experimentally deleted, there was a much lower response to chemotherapy.

"These data suggest that cells with low DUSP4 expression are enriched during NAC and that low DUSP4 expression in residual resected breast tumors is a potential biomarker for drug resistance and a high likelihood of tumor recurrence," said Balko.

The group also hypothesized that DUSP4 may be a potential biomarker for response to drugs that inhibit the MEK kinase. Using DUSP4-deficient tumors established in mice, they compared treatment with the chemotherapy drug docetaxel, with and without the MEK inhibitor selumetinib. This study showed that the combination was much more effective than docetaxel alone at eliminating the mouse tumors.

"These data support exploratory clinical trials combining chemotherapy and MEK inhibitors in patients with DUSP-deficient basal-like breast cancer," said Balko.

However, many patients still have residual cancer in the breast after neoadjuvant chemotherapy (NAC) is completed. These patients are at a higher risk of cancer recurrence and death.

The investigators suspected that profiling tumors after neoadjuvant chemotherapy would identify genes associated with resistance to this form of treatment. They studied gene expression patterns in 49 breast tumors obtained during surgery after four months of NAC.

They identified and analyzed specific groups of genes associated with high-grade, chemotherapy-resistant tumors, labeling their 244 unique genes the CLUSTER signature, and combined this panel with previously identified gene signatures to search for distinctive patterns of behavior.

The investigators found that low concentrations of dual specificity protein phosphatase 4 (DUSP4) is strongly correlated with faster tumor cell growth following neoadjuvant chemotherapy. Low DUSP4 was also correlated with a type of breast cancer known as basal-like breast cancer (BLBC). DUSP4 promoter methylation and gene expression patterns of Ras-ERK pathway activation were also higher in BLBC relative to other breast cancer subtypes.

When DUSP4 was present, chemotherapy was effective against cancer cells, whereas when DUSP4 was experimentally deleted, there was a much lower response to chemotherapy.

"These data suggest that cells with low DUSP4 expression are enriched during NAC and that low DUSP4 expression in residual resected breast tumors is a potential biomarker for drug resistance and a high likelihood of tumor recurrence," said Balko.

The group also hypothesized that DUSP4 may be a potential biomarker for response to drugs that inhibit the MEK kinase. Using DUSP4-deficient tumors established in mice, they compared treatment with the chemotherapy drug docetaxel, with and without the MEK inhibitor selumetinib. This study showed that the combination was much more effective than docetaxel alone at eliminating the mouse tumors.

"These data support exploratory clinical trials combining chemotherapy and MEK inhibitors in patients with DUSP-deficient basal-like breast cancer," said Balko.

Europe bailout of Spain could cost $125 billion

(AP) ? Spain became the fourth and largest country to ask Europe to rescue its failing banks, a bailout of up to ?100 billion ($125 billion) that leaders hoped would stabilize a financial crisis that threatens to break apart the 17-country eurozone.

The rescue offer follows growing pressure from international investors and the Obama administration and comes a week before elections in Greece, whose voters could decide whether the country leaves the euro.

Europe's widening recession and financial crisis has hurt companies and investors around the world. Providing a financial lifeline to Spanish banks is likely to relieve anxiety on the Spanish economy ? which is five times larger than Greece's ? and on markets concerned about the country's ability to pay its way.

"What the markets are looking for is essentially the Spanish government's acceptance that its banks are broke," said Jacob Kirkegaard, a research fellow at the Peterson Institute for International Economics in Washington, Saturday.

Economy Minister Luis de Guindos announced the deal after an emergency conference call with eurozone financial leaders. He said the aid will go to the banking sector only and would not come with new austerity conditions attached for the economy in general ? conditions that have been an integral part of previous bailouts to Portugal, Ireland and Greece.

The exact figure of the bailout has not yet been decided. De Guindos said the country is waiting until independent audits of the country's banking sector have been carried out before asking for a specific amount. The audits are expected June 21 at the latest.

De Guindos did say, however, that Spain would request enough money for recapitalization, plus a safety margin that will be "significant."

With markets in turmoil, de Guindos said the government's efforts to shore up the financial sector "must be completed with the necessary resources to finance the needs of recapitalization."

Finance ministers of the 17 countries that use the euro said the money would be fed directly into a fund Spain set up to recapitalize its banks, but underscored that the Spanish government is ultimately responsible for the loan.

Still, that plan allows Spain to avoid making the onerous commitments that Greece, Ireland and Portugal were forced to when they sought their rescues. Instead, the eurogroup statement said that it expected Spain's banking sector to implement reforms and that Spain would be held to its previous commitments to reform its labor market and manage its deficit.

The eurogroup statement said that meant the cost could reach ?100 billion.

The Spanish acceptance of aid for its banks is a big embarrassment for Prime Minister Mariano Rajoy, who insisted just 10 days ago that the banking sector would not need a bailout. He was elected in November and walked right into a hurricane.

International pressure on Spain to solve its financial problems has grown more urgent in recent weeks. On Thursday ratings agency Fitch hit Spain with a three-notch downgrade of its credit rating. That left it two levels above junk status. Then on Friday, Moody's Investor Services warned it could downgrade Spain and other countries in the eurozone.

The International Monetary Fund early Saturday released a report estimating that Spanish banks need a recapitalization injection of at least ?40 billion ($50 billion) following a stress test it performed on the country's financial sector. That report came out three days ahead of schedule, underscoring the urgency of the situation.

And U.S. President Barack Obama, facing re-election, enduring a weak economy and in need of strong trading partners, expressed strong concern late Friday over the European economic crisis.

U.S Treasury Secretary Timothy Geithner welcomed Spain's decision and the offer of European support, describing them as "important for the health of Spain's economy and as concrete steps on the path to financial union, which is vital to the resilience of the euro area."

French Finance Minister Pierre Moscovici said the deal would "contribute to restoring confidence in the eurozone."

"The accord announced tonight speaks to a reinforced solidary among the countries of the eurozone and to their resolute desire to ensure its stability," he said in a statement.

Spain's financial problems are not due to Greek-style government over-spending. The country's banks got caught up in the collapse of a real estate bubble. However, as Spain's leaders have struggled for a solution to their banking crisis, the country's borrowing costs have soared close to the level that forced the governments of Greece, Portugal and Ireland to seek rescues.

Some of Spain's banks are struggling with by toxic real estate loans and assets. The Bank of Spain says they total around ?180 billion. Nationalized lender Bankia, SA, which has requested ?19 billion in aid, has ?32 billion in toxic assets. Around four other banks are considered prime candidates for bailouts. De Guindos said Saturday the sector is largely solid and the euro zone package will be funnel toward only about 30 percent of it.

Analyst Rafael Pampillon if IE Business School in Madrid said the bailout addressed the uncertainty the markets had felt about how Spain's debt-laden banking sector would recapitalize.

"This uncertainty, and hence the panic, will slowly dissipate from the markets," he said. Pampillon added that with polls forecasting a pro-Euro victory in Greek elections, markets would be further relieved because the austerity conditions imposed on Greece would most likely be fulfilled.

Eswar Prasad, a Cornell University trade policy professor and senior fellow at the Brookings Institution, said the decision "buys some temporary breathing room for the eurozone."

Moody's said Spain's banking problem is largely confined to that country and not likely to spill over to other eurozone nations, with the exception of Italy ? where the European Central Bank has already stepped in to buy government bonds as a way to help lower the country's borrowing costs.

Spain has been criticized for being too slow to set out a roadmap to resolve its problem. European business leaders and analysts have stressed that Spain must find a solution quickly so that it is not caught up in any market turmoil sparked by the June 17 Greek elections. There are concerns that anti-bailout left-wing party Syriza could become the largest party in the Greek parliament, putting the country's membership in the eurozone at risk.

Working in Spain's favor is the fact that its public debt is actually quite low, at 68.5 percent of its gross domestic product at the end of 2011.

Its debt is predicted to hit 78 percent by the end of the year, but even that figure would be below the debt-to-GDP ratios of Europe's strongest economy, Germany, which is at 82 percent.

But Spain's in its second recession in three years, with unemployment at nearly 25 percent and little hope for improvement this year. Prime Minister Mariano Rajoy's government has imposed a wave of austerity measures since he took office in December that have raised taxes, made it cheaper to hire and fire workers and cut government funding for education and health care.
